Abstract本发明涉及一种双脂膜表面改性的蛋白质芯片,其以硅基片为固体基片,其上依次为与硅基片共价键结合的L-α-磷脂酰乙醇胺单分子层,和与该磷脂酰乙醇胺单分子层通过疏水作用结合的对硝基苯酯基-聚乙二醇-(1,2-双油酰基-3-甘油磷脂乙醇胺)层。该蛋白质芯片是在微流道系统中将L-α-磷脂酰乙醇胺单分子层共价固定到硅基片表面,然后吸附对硝基苯酯基-聚乙二醇-(1,2-双油酰基-3-甘油磷脂乙醇胺)得到的。该蛋白质芯片可以用于椭偏成像生物传感器,通过在该蛋白质芯片表面PEG的末端官能基来共价固定配基分子,探测能与其特异性结合的靶分子,二者结合后,在椭偏成像系统中可以观察到蛋白质芯片表面形貌变化。本发明的双脂膜表面改性的蛋白质芯片可以有效阻止非特异性吸附,其配基分子层具有很好的稳定性,且保持高生物活性。
